Description
Hormone releasing factors and releasing hormones are signaling molecules produced by glands in multicellular organisms. The glands that secrete Luteinizing hormones LHRG and LH, FSH comprise the endocrine signaling system. The term growth hormone releasing hormone GHRH is sometimes extended to include chemicals produced by cells that affect the same cell (autocrine or intracrine signaling) or nearby cells (paracrine signaling). Human recombinant LHRG and GHRH are produced in E. coli or in yeast cells.A microtiter plate (spelled Microtiter is a registered trade name in the United States) or microplate or micro well plate or multiwell, is a flat plate with multiple "wells" used as small test tubes. The microplate has become a standard tool in analytical research and clinical diagnostic testing laboratories. A very common usage is in the enzyme-linked immunosorbent assay (ELISA), the basis of most modern medical diagnostic testing in humans and animals. A microplate typically has 6, 24, 96, 384 or 1536 sample wells arranged in a 23 rectangular matrix. Some microplates have even been manufactured with 3456 or 9600 wells, and an "array tape" product has been developed that provides a continuous strip of microplates embossed on a flexible plastic tape.The receptors are ligand binding factors of type 1, 2 or 3 and protein-molecules that receive chemical-signals from outside a cell. When such chemical-signals couple or bind to a receptor, they cause some form of cellular/tissue-response, e.g. a change in the electrical-activity of a cell. In this sense, am olfactory receptor is a protein-molecule that recognizes and responds to endogenous-chemical signals, chemokinesor cytokines e.g. an acetylcholine-receptor recognizes and responds to its endogenous-ligand, acetylcholine. However, sometimes in pharmacology, the term is also used to include other proteins that are drug-targets, such as enzymes, transporters and ion-channels.
Assay principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Parathyroid Hormone Receptor 1 (PTHR1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Parathyroid Hormone Receptor 1 (PTHR1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Parathyroid Hormone Receptor 1 (PTHR1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Parathyroid Hormone Receptor 1 (PTHR1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ELISA's stability

It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
Precision of the test
Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Parathyroid Hormone Receptor 1 (PTHR1) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Parathyroid Hormone Receptor 1 (PTHR1) were tested on 3 different plates, 8 replicates in each plate. ELISA's specificity
This assay has high sensitivity and excellent specificity for detection of Parathyroid Hormone Receptor 1 (PTHR1). No significant cross-reactivity or interference between Parathyroid Hormone Receptor 1 (PTHR1) and analogues was observed.
Warnings
The Stop Solution is acidic. Do not allow to contact skin or eyes. Calibrators, controls and specimen samples should be assayed in duplicate. Once the procedure has been started, all steps should be completed without interruption.
Storage recommendation
-20°C. Bring all reagents to room temperature before beginning test. The kit may be stored at 4°C for immediate use within two days upon arrival. Reseal any unused strips with desiccant pack. Minimize freeze/thaw cycles.
